OpGen Creates Map of World’s Most Lethal Fungus

Madison, Wis.-- The world’s most common lethal fungal infection is caused by a mold called Aspergillus fumigatus. Strains put transplant, leukemia and HIV/AIDS patients at serious risk. OpGen, a University of Wisconsin-Madison spinout, has created a whole genome map of this fungal pathogen that will help a consortium of international researchers to find new therapeutics to treat this, and other deadly infections. The company believes their technology will lead to major breakthroughs in medical research, diagnosis and treatment.

Research on this source of infection dates back to England in 1842 and the first fatal infection was described in 1953. Less serious strains of the mold have been identified as the cause of allergies and sinus infections. OpGen’s DNA optical analysis technology generated massive volumes of data that will allow scientific and medical researchers from the United Kingdom, United States, France and Spain to study the complete genome sequence of this organism at a greatly increased speed and reduced cost.

“OpGen’s whole genome map gave us the ability to put everything together, allowing our team to view all of the pieces of the chromosomes as a whole unit,” said William Nierman, consortium member and investigator with The Institute for Genome Research. “Because of the nature of this global project, we thought we would have to expend a great deal of money trying to link all the parts together and were faced with the possibility that it might not have worked. With OpGen’s help, we were able to link all of the sequence contigs into whole chromosomes in a very cost efficient manner.”

According to Colin Dykes, chief scientific officer of OpGen, the creation of an optical genome map is an alternative to a costly and time consuming cloning strategy that could takes months.

“All the data was collected on a Friday from a DNA sample, captured on specialized glass chips, and processed over a weekend. We created 300 different over-lapping maps which reduces errors,” Dykes said.

Professor David Schwartz, OpGen’s founder, developed the technology along with his scientific team at UW-Madison. “This is equivalent to mapping the human genome three times over,” Schwartz said.

Breakthrough will lead to personalized medicine

The current commercial application of OpGen’s technology follows the company’s December 2003 announcement of a genome database that could be used to identify biological agents that could be used for bioterrorist attacks.

According to Dykes, OpGen’s technology can also be used to study the history of a disease and analyze the genomic differences. He believes that optical DNA mapping is a step toward human genome analysis and that it proves the value of genome sequencing technology, which will lead to breakthroughs in medical technologies and treatments.

“We are well on the way to achieving the capacity to perform genome mapping in large human populations” Dykes said. “We will be able to compare many genomes to discover why some people respond differently to medications and therapies. Adverse reactions to prescription drugs are reported to kill around 100,000 patients each year in the United States making it the sixth leading cause of death.This powerful technology will speed the development of personalized medicine."

"OpGen’s number one goal is to revolutionize pharmacogenomics and the development of novel diagnostics products,” Dykes added. “We are on track to perform our first human research studies by summer and commercial pharmacogenomic studies by the end of the year.”

I have had fungus in the sinus. My doctor removed the lining of my left sinus. I am now once again having some problems that I hope are not fungus. I am looking for a doctor to treat me. Any recommendations would be appreciated. I live in Salt Lake City Utah. Thanks much!

Three years ago I was diagnosed with colon cancer. Chemotherapy and radiation preceeded colon surgery and ten months later reconnection surgery. Prior to the cancer I was never sick, since then it seems I am never well. Cronic sinus infections, sore throat, swollen glands, low grade fever, bronchitias and itching in the right ear canal.

The general practicioner has me on a different antibiotic every week and has resorted to steriods. I am certain that all this has killed all my good bacteria along with the bad, causing yeast to develop into fungus in both my sinus cavities and my right ear canal and I cannot get a doctor to listen to me. They just keep putting me on more antibiotics.

I would like to purge my body of all the yeast and fungus. Is there any help out there?

I hope by now you have found that most holistic practioners, from chiropractor's, to naturopaths, to cleansing and detoxing clinics have been working with candida/yeast for years. Its done naturally with diet for starts.
Sometimes nystatin. Do a search on systemic candida remedies.

I AM SO GLAD TO HEAR THAT A STUDY MIGHT BE DONE. I HAVE BEEN FIGHTING SINUS INFECTIONS IN MY HEAD FOR ABOUT 5 YEARS NOW. I AM MISERABLE. I AM PUT ON 1 ANTIBIOTIC AFTER ANOTHER. I HAD STAPH. PUT ON BIAXIN FOR 21 DAYS. I NOW HAVE ASHMA, ALL KIND OF ALLERGIES AND A CONSTANT HEADACHE. MY BODY IS GIVING IN AGAIN. I HAVE PAINS IN MY CHEST, LUNGS AND BAD HEADACHE. I HAVE BEEN RUNNING A LOW GRADE FEVER FOR YEARS.I COULD NOT GET A DOCTOR TO TAKE ME SERIOUS. I KNOW THAT MY BODY IS BEING AFFECTED BY THIS DISEASE INTERNALLY. I HAVE RESORTED TO JUST GIVING UP. MY MOTHER IN LAW ALSO WAS FOUND TO HAVE STAPH. SHE WAS PUT ON VANCOMICIAN FOR 30 DAYS IV THERAPY. IT HAS SEEMED TO HELP HER, BUT HER VALVE IN HER HEART DOES NOT CLOSE PROPERLY. SHE HAS LUNG PROBLEMS ALSO. THIS STUDY IS REALLY NEEDED. I WOULD BE HAPPY TO PARTICIPATE, TO KEEP SOME ONE ELSE FROM GOING THRU WHAT I AM PRESENTLY ENDURING. I AM A GO GETTER. hi,
i now have aspergillus in my lungs and i'm alway sick.I got it from the air condition unit in the apts. i was living in.I am trying to get all the information about it that I can. i need all the information on ASPERGILLUS AND MOLD/MILDEW/FUNGUS IN AIR CONDITIONING UNITS IN THE HOUSE THAT I CAN GET AND IT'S LONG TERM AFFECTS THAT IT WILL HAVE.PLEASE PLEASE CAN ANYONE HEP ME!!!!!!!! Since starting work at a new , OLD school in July I have had one problem after another. I started by thoroughly cleaning my room in mid-July and went on vacation. I have had 3 emergency room visits since; each time - HIGH fever, EXTREME boddy ache/pain. For 5 months now I have continued to run a low grade fever and at time up to 103! My school building, on one particular wing of the building has had MULTIPLE sicknesses among those teachers and students. Mold/mildew is seen and KNOW (but NOT dealt with). The custodian was even threatened to be FIRED if he MENTIONED those words!
My doctor says this could not cause a fever. I have read otherwise. Can anyone help confirm this, fever thing, fatigue, muscle aches (continuous since this new job in this part of the building)? How can I have the building checked with TOTAL ANANEMITY for fear of being fired?

I had sinus problems for about 4 years. Tried everything from Allegra to all kinds of nasal
sprays and antibiotics. Nothing helped. After antibiotics I started having bladder infections
too. Finally I watched Doug Kauffman (TX)
program about fungus and chronic candida.
I bought one of his book (www.knowthecause.com)
and finally everything made sence for me.
I started taking grapefruit seed extract (www.nutribiotic.com) 20-30 drops in water or
juice between meals 3-4 times per day and I
feel 100% better. I do not have bladder infection any more, my sinuses dramatically improved and I
do not get colds. My husbands snore much less
and his sinus problems improved too. My dog
(poodle) had joint pain and ears inlammation, he could not walk and was on antibiotics and
strong pain reliever. Vet said he will have
to take medication for the rest of his life,
because there is no cure for his disease. Now
after taking grapefruit seed extract 3 times
per day (3 drops in his food or a treat: he is
12 pounds) for just 2 weeks, my dog running, playing and fully recovered.
I just need to say: I did cut sugar, yeast,
bread, corn, peanuts, hard cheeses out of my diet for 1 month. Also, I take probiotics and
caprylic acid to restore normal flora. The main thing for everyone with sinus problem is to help their immune system to recover from antibiotics
and gain its strength back. I hope this
information will help.

My husband has been running a low grade fever, fatigue, headaches and a sore neck and back off and on since Feb. 2007. The doctor took test and said he was healthy for his age, nothing wrong. Please help!
